摘要
Phenotypic 突變s are amino acid changes caused by mistranslation. How phenotypic 突變s affect the adaptive 演化 of new 蛋白質 functions is unknown. Here we evolve the 抗生素 resistance 蛋白質 TEM-1 towards resistance on the 抗生素 cefotaxime in an Escherichia coli strain with a high mistranslation rate. TEM-1 族群s evolved in such strains endow host cells with a general growth advantage, not only on cefotaxime but also on several other 抗生素s that ancestral TEM-1 had been unable to deactivate. High-throughput sequencing of TEM-1 族群s shows that this advantage is associated with a lower incidence of weakly deleterious genotypic 突變s. Our observations show that mistranslation is not just a source of noise that delays adaptive 演化. It could even facilitate adaptive 演化 by exacerbating the effects of deleterious 突變s an
